Development and Validation of a High Sensitivity On-Line Extraction LC-MS/MS Method for the Determination of Exemestane and its Metabolite 17▀-Hydroxyexemestane in Human Plasma

Summary Exemestane is an orally active irreversible steroidal aromatiase inhibitor used for the therapy of metastatic postmenopausal breast cancer.The objective of this work was to develop and validate a highly sensitive on-line extraction LC-MS/MS method for the determination of exemestane (EXE) and 17▀-hydroxy- exemestane (17▀ EXE) in human plasma.. A high sensitive, accurate and robust method for the determination of exemestane and its metabolite 17▀-OH exemestane was developed. The study also demonstrates how the use of selective analyte isolation techniques, chromatography and MS/MS detection make it possible to analyse steroidal compounds susceptible to isobaric interferences at trace levels.
